Mystery Queen revolves on the story of Yoo Seol-ok, a mystery novel loving ajumma who starts solving real life cases together with police detective Ha Wan-seung. Yoo Seol-ok always dreams of being a detective since she was a child but missed out on her chance to become a police officer after she married a prosecutor at a young age. At some point in her married life, she meets Ha Wan-seung and ends up fighting crimes and solving difficult cases with him. Although their approach in solving the cases are different, they turn out to be the perfect partners against crime mysteries.

Choi Kang-hee as Yoo Seol-ok. She always dreamed of becoming a detective but real life got in her way. She finds hope of fulfilling this dream because of an unsolved crime that leads her to partner with a Ha Wan-seung. Her experience in reading mystery novels helps her solve cases using theoretical knowledge and deductive reasoning.
Kwon Sang-woo as Ha Wan-seung. He is an ace violent crimes detective who graduated with honors from Police University. Unlike Yoo Seol-ok who relies on logic to figure out everything, he trusts his intuition and gut feeling while working in the field, making her team up with her an exciting crime-fighting duo.
Lee Won-geun as Hong Joon-oh. Hong Joon-oh is a rookie police officer who singles out Yoo Seol-ok to be the civilian consultant on a case he’s working on.
Shin Hyun-bin as Jung Ji-won. Jung Ji-won is an elite lawyer whose father is the owner of the largest law firm in Korea.
